Background:There is a paucity of empirical data regarding the effectiveness of Seroquel in the management of dementia and in improving cognitive functioning.
Methods:A systematic review was conducted of randomized controlled trials (RCTs) comparing Seroquel with placebo, or placebo alone or in combination with other treatments and in comparison with cognitive therapy in patients with dementia. The findings of the review were used as a basis for meta-analysis of clinical trials. We included all RCTs with a minimum size of 100 patients (n = 3060). The number of patients per protocol was determined using the “ratio” method.
Results:A total of 30 RCTs comparing Seroquel with placebo, or placebo alone or in combination with other treatments and in comparison with cognitive therapy were included in our review. In total, 608 patients (mean age, 60.8 years) met the inclusion criteria. The mean duration of Seroquel use was 17.8 months (standard deviation [SD], 7.5 months). In comparison with placebo, the mean duration of Seroquel use was significantly more frequent in patients who received cognitive therapy (mean [SD] = 1.7 months) than in patients who received cognitive therapy alone (mean [SD] = 1.3 months).
Conclusion:The results of this systematic review showed that Seroquel may be more effective than placebo in improving cognitive functioning in patients with dementia. However, more rigorous trials are needed to evaluate the benefit of Seroquel in such patients.
